Valdoxan (Agomelatine) Combinations

Posted by beyondcrazy on December 28, 2010, at 7:14:07

Hello.

Im interested in people taking Valdoxan (Agomelatine) with other meds.

Because its a new drug my Doc is hesitant about trying too many meds with Valdoxan.

My combo was Dexamphetamine and Valdoxan with no interactions.

I would like to try Cymbalta and Valdoxan.

Anyone here on that combo?
What other meds have you used with Valdoxan?

Thanks in advance.

BC.

My doctor prescribed Valdoxan to me a while back. I ended up on the top dose (50mg?). I'm afraid I didn't find it that effective as an antidepressant, although it did help me sleep. While on the drug, I was also taking (per day):

600mg lithium carbonate
600mg Lyrica (pregabalin)
40mg buspirone

From what i know of Valdoxan it ie a relatively safe drug, however, a search for drug interactions on the net may be useful (if you haven't already carried out one).

There is something about the CYP enzyme thing, which of course I don't understand.

Anyway, for that reason it is hazardous to combine it with Serzone. I really don't know, I'm just trying to remember.

I have heard of people combining it with low dose SSRIs safely.

BC,

> I would like to try Cymbalta and Valdoxan.

The use of Cymbalta and Valdoxan together is contraindicated (i.e.; a no-no) because both meds are metabolized by cytochrome P450 1A2 (CYP1A2).

Here is a link to the Valdoxan Prescribing Information Document:

http://www.servier.co.uk/pdfs/Valdoxan_SPC.pdf

Read Sections: 4.3, 4.4 Subsection - Combination with CYP1A2 inhibitors, and 4.5.

Here is a link to a more comprehensive Valdoxan Prescribing Information Document and other Valdoxan information. Click the "Doctor" option to access the most comprehensive information.

http://www.valdoxan.co.uk/landing.aspx?redirect=/Default.aspx

Here is a link to the Cymbalta Prescribing Information Document:

http://pi.lilly.com/us/cymbalta-pi.pdf

Read Sections: 5.10, 7, 7.1, 7.2, 7.8, and 12.3.

Discuss this CYP1A2 issue with your p-doc. The bottom line is that since both Valdoxan and Cymbalta are metabolized by CYP1A2, the blood levels of one or both will likely be altered from what would ordinarily be expected from the oral dosages.

I take 12.5 mg/night of Valdoxan and it stopped my bipolar II ultra rapid cycling and bipolar II depression, when nothing else ever has.

Below is a list of what I take.

-- Ron

dx: Bipolar II and mild OCPD

600 mg/day Trileptal (oxcarbazepine)
200 mg/day Lamictal (lamotrigine)
500 mg/day Keppra (levetiracetam)
90 mg/day Nardil (phenelzine)
12.5 mg at betime Valdoxan (agomelatine)
3.75 mg/day Deplin (taken with 2500 mcg/day of sublingual methyl B-12, and 12.5mg/day of sublingual P-5-P)
2.5 mg/day Adderal XR

45 ml/day of Carlsons Bottled Fish Oil
100 mg/day phosphatidylserene
1 Centrum Chewable Multi-vitamin/day; Only 100% of all the usual vitamins; NO VANADIUM!!
2000 IU/day Vitamin D-3
850 mg/day of Mg 212% of RDA (as 5 grams of Mg Malate)
200 mcg/day GTF Chromium
600 mcg/day Chromium Picolinate
200 mg/day Co-Q10
1000 mg/day Cinnamon
480 mg/day Milk Thistle

I take Valdoxan with Cymbalta. I have no idea how that gels with my liver.
