Not literally a protein. It's a modified amino acid, which requires serotonin as its raw material. It's four steps removed from l-tryptophan.

Stomach, with acid and enzymes.

It gets into the brain in useful amounts. There are a number of processes that get some of it, along the way, but some makes it all the way.
