Tell me about Ondansetron for augmentation

Posted by myco on May 3, 2009, at 23:42:47

I read a study recently demonstrating that Ondansetron (a 5ht3 antagonist) reversed the dopamine firing rate attenuation that Clorgiline had in rats. Clorgiline being specific for MAO-A.

I'm looking at augments to prevent/slow the dopamine firing rate attenuation that is so common with long term MAOI use. My first try is gonna be Memantine. Perhaps Ondansetron second.
Also considering Pramipexole...but that one might send me through the roof with anxiety...dunno.

Would a D2/D3 agonist like pramipexole resolve your issues with dopamine firing rate? Just interested in how that would work.

No I dont suspect it would. It would simply just give a boost here and there to DA. I would probably end up going pseudo-bipolar again, like when I overdid L-Arginine, if I took it too much.

Isn't that an anti-nausea drug?

yes it is. it's a 5HT3 antagonist...has many potential uses apart from antinausea (i.e. as an AD and augment)

It has been used experimentally in psychiatry.

Yes it seems that since its patent has expired, generics from the former Rite-Aid owned pharmacy drugstore.com for ondansetron as it is I think the oldest 5HT3 blocker, have gone down to about $1 a pill (8mg). I'm not going to get into shady 'offshore' pharmacies for obvious reasons.

There are about 8 or 10 generic companies looking at the FDA's orange book.

5HT3 medications though such as Kytril (granisetron) are still prohibitively expensive even with a generic out, except for the cost-benefit decision for their intended purpose of short term chemotherapy rounds with nausea.

It is also thought that ginger is a weak 5HT3 blocker from some studies.
