Agomelatine Approved in EU

Posted by Netch on February 25, 2009, at 9:22:52

Agomelatine Approved in EU for Major Depressive Episodes:
The European Commission has granted marketing authorisation for agomelatine (Valdoxan) for the treatment of adult patients with major depressive episodes.

The approval is based on both short-term and long-term results from a large, comprehensive, international development programme including almost 6,000 adult patients with depression.

The results demonstrated the superior efficacy of agomelatine as compared with placebo, selective serotonin reuptake inhibitors (SSRIs), and serotonin noradrenaline reuptake inhibitor (SNRI) treatments.

Agomelatine was effective in moderately and severely depressed adult patients (18-65 years) presenting a first or recurrent episode of depression. Clinical improvement was seen within the first week of treatment, as reported by physicians and patients.

The drug was effective against all the core symptoms of depression including depressed mood, anxiety, feeling of guilt, psychomotor retardation, sleep disturbances, and daytime fatigue.

I will probably write a very short report/note about how I experienced it.. could be usefull for some of you out there who like me gets all kinds of weird effects from substances affecting the brain. Im not gonna bet all my cards on this one but remembering back to the few times melatonin worked it sure would be amazing with a blast from the past :P

I've read some have privately imported agomelatine from Ukraine where it's already available. Their experiences seem to be mixed. Some seem to embrace it others not.

Medical trials show that the success rate of Agomelatine is about the same as other AD but with lower side effects and improved sleep.

I wonder what happens if you take valdoxan some time during the day (as opposed to taking it before bedtime which you should do according to guidelines)? A melatonin-effect during the day... could that somehow be beneficial or just the opposite? Any ideas from someone?

Hi,

Denise here from the uk, I used to post here a lot but have only lurked lately.

Please tell me that this drug won't just be like Remeron (I didn't respond well to) along with Melatonin as I've been having a really bad time lately with my depression and anxiety and looking for some hope.

Does anyone know?

In what way is it different or any better?

How do you know?

I took Remeron twice, and twice only.
I'm not in a position where I can take stuff like that.
Agomelatine feels benign, and distinguishes itself among psychiatric drugs by not feeling like a pharmacological insult.

So, you are going to ask, has it helped?

Yes.

How?

I find social situations much easier to deal with....I'm calmer, and can process the info better. I feel less vulnerable and feel like I have more skin around me. I've only been taking it for a couple of weeks.

Too soon to tell, but I don't notice an increased desire to eat.

Sigismund,

I didn't realise that you were on it. I'm not sure what it is you suffer from but does the drug seem to be helping at all?

Sorry, ignore my last post, I've just seen your response about how you are feeling on it.

What oher meds have you tried in the past and how have they affected you?

I suffer from chronic anxiety (not social anxiety or phobias). Do you think it might help with that?

>What oher meds have you tried in the past and how have they affected you?

Not SSRIs or TCAs
Benzos
Opiates
Melleril/Stellazine/Anatensol and the whole horrible bag.
The popular drugs were (naturally) the ones I enjoyed most.

>I suffer from chronic anxiety (not social anxiety or phobias).

Well, I don't suffer from much anxiety as such (in seclusion).
Some (most?) people just give me the jerks, or I give myself the jerks around some people.
I take this to be a problem I have in processing social information into a set of rules I can follow, as I have no instinct for life :)

>Do you think it might help with that?

There is an agomelatine anxiety thing (in the 6 hours before the evening dose perhaps), which I do not understand properly yet.

The really good thing about agomelatine is that it does not feel just bloody awful.
My first response was 'this is the psychotonic thing'.
I've always needed a tonic.

Don't feel put off though.

Yes and no.

To begin with it *was* calming (and clean).

After a couple of weeks I found myself, in the afternoon, feeling like I really really needed a drink and a Valium, so I have halved the dose for the moment.

Hi Denise, I have GAD and have been taking Agomelatine for about 4 months now. I started on the regular dose 25mg, and have since gone up to 50mg. The drug does have a good antidepressant anti-anxiety effect. However I think most people will need to take the higher dose of 50mg to get the most out of this drug. 25mg was ok but still had some anxiety symptoms (no panic attacks), 50mg has been a marked improvement on that, I think I may reach remission in the coming months.

Ago is much better than SSRI's for side effects. No/minimal sexual dysfunction, no increased appetite and much more balanced emotional state.

So far I'm very happy to be taking it.

Negcreep :)
