I've heard from various sourses that it is a dopamine reptake inhibitor, thats why it is used for Stimulant Tolerance, then i read in a post that some one accidently took it with Nardil, and said it was a serotonin repake inhibitor, which caused serotonin syndrowme.

Dextromethorphan inhibits the reuptake of serotonin but not dopamine. Several years ago, there was some speculation that DXM might inhibit the dopamine transporter; however, this claim was never substantiated. See http://www.ncbi.nlm.nih.gov/entrez/query.fcgi?cmd=Retrieve&db=PubMed&list_uids=9105700&dopt=Abstract for evidence that suggests that DXM inhibits the serotonin transporter. DXM is also an NMDA antagonist, a sigma-1 receptor agonist, and a sigma-2 receptor agonist. It binds with less affinity to the NMDA channel and to sigma-2 receptors. Its metabolite, dextrorphan, is also an NMDA antagonist and a serotonin reuptake inhibitor.

I wonder if all that DXM for stimulant tolernace is really just a myth, i dont know. I understood why if it was a dopamine reuptake inhibitor, increase dopamine levels to keep the same effect going for amphetamine or whatever.
I wonder if you were depressed, it would be a good mood elevtor. I heard too much is VERY unpleasant.

Hi Shawn.
What are some of the biological functions of the sigma receptors?
Thanks (as usual).

iv been trying to find the sorces but i just cant seem to get them again. all i know is that when i am on DXM (spiritual doses) i can just, not smoke. a pack and a half a day,ex-heroin addict, nerotic smoker just on a whim decides to throw out his ciggeretts. anyway i had very mild cravings and eneded up quiting for like a month, but when i tried quiting when i wasnt on DXM i couldnt do it all, it was damn neer imposable. anyway i started smoking for like a weekend (back up to a pack for the three days) and i quit again a few days ago. these things are horribly addictive. As for DXM there is just not enough research done on it, its more of medical enigma.
anyway ,
peace j(p.s. it does chill out my smack feinds, but it is no replacement for methadone)

One theory is that sigma-1 receptors are intracellular amplifiers for signal transduction (Su and Hayashi 2003). DHEA is a sigma-1 agonist.
Sigma-2 receptors may serve a similar role; they are known to modulate neurotransmitter release.Shawn
