its a long time before they're out (if they come out)i'm sure but one is in clinical trials at least right now.

there are three new meds being worked on right now. a new Class of AD called SNDRI (serotonin, norepinephrine DOPAMINE reuptake inhibitors)...just what 'I' am looking for.

the three i found are called:
tesofensine (currently in clnical trials)
brasofensine (under development)
and
GlaxoSmithKline's NS2359 (under development)

however Tesofensine will most likely be marketed as a treatment for obesity (as reserached in phase II trials) now they are in phase III but it didn't say what for, if they've switched to look at potentially depression/bipolar or staying with appetite suppressant. it was originally researched for treatment of alzheimer's and parkinson's.

Brasofensine was researched for treatment of parkinson's and depression. the bad news is Phase II trials were conducted in 1996 and though was shown to be well tolerated the current status of this drug is unknown.

and i couldn't find any info on the GSK drug, so i don't know the status of this.

i'm writing about these meds incase anyone out there has more information about these meds.

i think the key to my issue is dopamine and if i could find a different drug that targets dopamine i'm thinking specificially D1 it could really help.

Brasofensine is no longer in trials. Its been closed down.

Tesofensine will start phase 3 trials in obesity around yearend

And NS2359 is currently in 2 big phase 2 trials for depression under the name gsk372475. Results is due in 6 month time

-any indication for ADD/HD, Narcolepsy?

MMMM a 5ht, Dop, Norp reuptake inhibitor? any releasing affinity for these?

Sounds similar to Desoxyns (M-amphetamine) MOA, which is also indicated for obesity.

Are these compounds related to nomifensine?

here's a webpage i found a long time ago. not sure how accurate it still is. but its got a listing of drugs in the works for various disorders (depression, anxiety, sleep, bipolar...)

sorry for posting so much but i searched that website i just posted and realized that it is pretty old and doesn't seem to have been updated.

it shows mirapex as phase II which has been out for a while now, also a new one Valdoxan, which i think i've heard a few people here mention already
(5-HT2C antagonist, f-HT2B antagonist, melatonin M1/M2 receptor agonist)

then there are two others that i would like to know more about if others have heard more of. they were specifically listed under bipolar. (psychosis) but also used for schizophrenia i believe.

zimaril (iloperidone)
(selective DA/NE/5-HT antagonist) at the time it was NDA submitted, which is one step away from being approved (though it would still take forever)

the second is
Asenapine (ORG 5222)
5-HT2 antagonst, D2partial agonist
also NDA submitted
though this one looked a lot at schizophrenia and i think bipolar mania more so than depression.

I'm afraid that link is going to be way over most people's heads on here. What was your point? Are you recommending Tesofensine as an anti-depressant because it works on all three major neurotransmitters. What is the drug currently used for?

The link was just a reply to the question about affinity for Tesofensine.

Although there also should be some quiet compelling preclinical data there suggest using Tesofensine as an anti depressant, it will not be developed in that direction. The focus is on obesity and possible also type 2 diabetes. But it is still 3-4 years from the marked.

NS2359/GSK372475 on the other hand is developed for depression. And it works more or less in a similar way as Tesofensine by altering the 3 neurotransmitters dopamine, serotonin and noradrenaline. The affinity for NS2359 is

Dopamine 8nm
Noradrenaline 0,6nm
Serotonin 5nm

GlaxoSmithKline plans to start several large phase 3 depression trials with NS2359/GSK372475 in the middle of 09. And they think that this new mechanism will be the next new big anti depression drug class and thereby overtake the SSRI class. Only time will show if thats true.

thanks i was looking for this info.
if all runs well, when do you think theyll try to get it on the market? 2010/11? or later

i really have my hopes for this drug since it has more effect with dopamine and with me this seems to be key.
and maybe, just maybe with this in mind, i can hang in there a while longer.

We have a SNDRI already, by taking Welbutrin with an SSRI or Effexor
