I hate to confuse you, but are you sure that you gave Parnate an adequate trial? I see too many people give up on a treatment after only a few weeks. It is not unusual for someone to experience ups and downs early in treatment. The ascent to remission is seldom a smooth one. It is a good sign that you experienced an antidepressant effect so early in treatment. However, it is not necessarily a bad sign that it did not continue uninterrupted. I think you need to give each dosage increase three weeks before evaluating its effectiveness. With Parnate, I don't think one can consider it a treatment failure until a dosage of 80mg is reached and given a minimum of 3 weeks to work.

Roche tried to get moclobemide approved in the US to treat social phobia in the 1990s. Roche felt that it had a better chance of having it approved for that indication since there were no drugs yet approved for that disorder. The studies to be submitted to the FDA did not show sufficient efficacy, and the drug was withdrawn from consideration.

Moclobemide tends to produce improvements in depression during the first two weeks of treatment at low dosages (150-300mg). Unfortunately, it usually becomes necessary to continually increase the dosage in order to maintain an antidepressant response. One can safely go up to 1200mg, but it doesn't usually make much difference.

Moclobemide was popular when it was first marketed because of its reversibility and favorable side effect profile. It was the most widely prescribed antidepressant in some countries for several years. However, its popularity declined quickly once its inferiority to other agents became apparent.

Of course, some people are very happy with moclobemide. It can work. However, I would place more confidence in the probability that the average person will respond more favorably to 80mg of Parnate than to 1200mg of moclobemide.
