Clomipramine is much more powerful as an NRI than Effexor, via its desmethyl metabolite, which is about equipotent to nortriptyline as an NRI.

Clomipramine is also more powerful as an SRI, but probably Effexor's low affinities for the NE and serotonin transporters are offset by its low level of protein binding.

A friend of mine who is currently on effexor told me clomipramine was far and away the most powerful AD he had ever taken. I would take it myself, if I could persuade my pdoc to give me a trial.
