Clomipramine For Social Anxiety

Posted by farshad on August 28, 2018, at 16:13:23

Hello, after looking around for a while, I have found a drug I think would greatly benefit my social anxiety.
Mainly becuase it reduces CRH (corticotropin-releasing-hormone).
Anyhow has anyone had success with this for social anxiety?

questions about studies:

According to this study: https://www.ncbi.nlm.nih.gov/pubmed/15214866

1. ''It was found that clomipramine lowers CRH mRNA expression in the PVN by 74%''
Lowers MRNA expression? is that the same as decreasing the activity of CRH?
the study was done on syrian hamsters but supposedly they are similar to humans so it would work the same. I wonder why they used the term ''lower'' , in my mind lower is usually used to describe height etc, why they didnt use decrease MRNA expression, just curious.

2. according to this study
https://www.ncbi.nlm.nih.gov/pubmed/11526469
''st johns wort given daily by gavage for 8 weeks but not for 2 weeks significantly decreased levels of corticotropin-releasing hormone (CRH) mRNA by 16-22% in the hypothalamic paraventricular nucleus (PVN''
So would combining clomipramine + st johns worth = decrease CRH activity up to 96%?

3. this study:http://psycnet.apa.org/record/1995-10569-001
''After beginning treatment with clomipramine, there was a significant decrease in CSF levels of CRH''

what is cerebrospinal fluid is it connected to the hypothalamus PVN and will decreasing CRH there affect your mood?

CRH aside, clomipramine is usually anxiolytic (after a few weeks of treatment, at least).

You may find it useful.

what other anxiolytic do you know about?

I have tried:
SSRI/SNRI
MAOI(Nardil)
benzo like xanax work of course but not a long term solution.
tried lyrica and related drugs didnt work.
I imagine most antipsychotic are anxylotic, I tried zyprexa once and that worked best for my anxiety.

All I know is I tried Clomipramine once and it was such a dirty drug with strong side effects that I think it would work really well for social anxiety amongst elephants. It's that strong. :-) Honestly I think the social anxiety benefit from it is probably due to the very strong antihistamine effect more than anything else. The serotonin stuff and norepinephrine stuff helps.

I can think of a handful of cheap over the counter herbal capsules that work better on social anxiety than clomipramine, and at a fraction of the cost and with near zero side effects.

Sometimes I think it might make more sense for someone to deal with their social anxiety in other ways most of the time - relaxation methods, yoga, herbs, supplements, deep breathing, exercise, whatever helps - but use a specific drug for special occasions rather than 24/7. For example if you're heading out to a dinner party and you're real nervous about it, there is no need to be hooked on some heavy duty med every day of your life for that. You can manage the rest of it.

But for those crucial times, there is always stuff like Xanax, Valium, Ritalin, Adderall, and others that can put you comfortably in the middle of a crowd.

Dirty is nice, isn't it?

antihistamine effect isn't anxiolytic.
Most probably it is the combination of strong SRI + anticholinergic effects + 5HT antagonism.

Or.... it is the CRH inhibition effect... which sounds the most plausible imo.

DNA > RNA > protein

clomipramine lowers mRNA so less CRH is made.
