5--ht2c-antagonistic effects of Fluoxetine(prozac)

(I'll just copy my post on imminst to this board for more answers and discussions, I think it's an interesting idea and want to see if anyone else has tried/thought about it...)


I have recently started taking the new AD agomelatine which is an mixed melatonin-agonist and 5-ht2c-antagonist.
Antagonism of the 5-ht2c-receptor seems to increase prefrontal levels of dopamine and noradrenaline and has anxiolytic effects.
I heard that at low doses, fluoxetine (aka prozac) has antagonistic properties at the 5-ht2c-receptor, well before the serotonin transporter is affected.

Does anybody know how low dosage you should take to benefit from this effect while avoiding any effect on the SERT-transporter?

I was thinking about combining Agomelatine (pro-sleep+5ht2c-antagonism), with selegiline (low dose to avoid MAO-A-inhibition) and fluoxetine for added 5ht2c-antagonism.

all i no is you dont take 5htp with antidepressants

I *think* Brainbeard posted somewhere that this could be done with 5mg/d of Prozac.

Perhaps he will turn up here.

I have more questions about this...Brainbeard, are you able to answer some of these questions?

I too am interested in maybe taking Prozac at a dose so low that it dose not inhibit the reuptake of serotonin. I want to do this in hopes that it would still antagonize the 5ht2c receptor and increase levels of dopamine and norepinephrine.

If a very low dose of Prozac acts this way, would it not be safe to combine such a low dose of Prozac with Zoloft? Then one could benefit from both the unique mechanisms of both right?

One more question...Anyone know of any medications other than the atypicals, that antagonize the D2 receptor?

Wait, won't prozac inhibit the reuptake of serotonin at any dosage? Or, even if it does not act as an SSRI at a very low dose initially, won't it eventually.

If a 2.5 mg dose of prozac does not act as an SSRI at any time, I would love to see how it might effect me while I am taking Zoloft. I don't see how there would be any guarantees with this Prozac 5ht2c antagonism without the SSRI effect.

I was once on paxil 7.5mg with luvox 12.5 and the benzos low doses of all meds. Phillipa

Huh really? And this was your doctor that prescribed this? Were you transferring from one medication to another? So I guess you had no problems.

I am just thinking that I can maybe get the benefits of prozac and Zoloft by taking a high dose of Zoloft, 100 to 150 mgs, and a low dose of Prozac, 2.5 to 5 mgs. Maybe this is a bad idea

No seriously was the pdoc very low doses of meds was in the hospital at the time for a month really sick did have lyme's disease. Pic line rocephin and then oral antibiotics pulsed for two years. Was also going through menopause at the time. Not pleasant at all. I just couldn't handle any med in high doses now that is when benzos really relaxed me didn't put me to sleep. Love Phillipa
