Phase III testing is still ongoing here in the States. Novartis has been delayed in finishing these trials. Not sure what this means about what sort of trial results they are achieving. Suffice it to say though that we have at least a year and a half or more should Novartis submit a NDA to the FDA this year (which now seems somewhat doubtful). The slightly good news is that it is now being launched in the wider EU this summer.

JB,
I live in Israel- do you know when it will be released here? We are not part of the EU.

I hope it is released in the US as well.

thanks

Israel has some EU economic ties but yes, not part of the EU.

As a side comment, being Jewish, some day I'd like to visit Israel, but I think that's a topic for social or religion, not that I am all that religious particularly.

I was reading some fairly recent posts by Marty. He said he orders Agomelatine by mail from some other country(not sure which country).--I understand this site does not permit people to list sources where they may order medications, however if its a reliable outfit, maybe an exception should be made in this case. And this is coming from a guy(me) that never ordered any medications online,I personally am not comfortable with the idea, but that's me. A sidenote:It's absurd that this medication and certain other possibly effective proven medications are not available in the US and some other countries yet.Why wait, they have been approved in other countries?---The FDA is terrible. If a medication has proven results it should be immediately available in every country.

Oh I don't know -- is that terribly wise, I mean EMEA and Canada's Health Products and Food Branch of Health Canada approve medications; India's health ministry, Japan, etc...

But are each created alike...no. Is it suffice to say that medications that are available in first class Western democracies are likely to be safe for other individuals, probably...

....but it goes much beyond that, including litigation which would be unaffordable to people injured unjustly by medications in developing countries just because a major health organization put a stamp on something.

And just like the FDA, EMEA (or corresponding countries) have had to recall things, such as the 'first' SSRI.

A well regulated health agency collects post-marketing data to see what happens in the real patient-hours after a medication has been approved. This can't be collected reliably from other countries. This is what is known here (FDA) as "Phase IV".

I am one of the people like Marty who has been ordering Agomelatine from the Ukraine for 6 months now. Works fairly well for my GAD and depression and little to no side effects.

Its difficult to tell you how to go about obtaining agomelatine without breaking the rules of this forum.

But I can always suggest to you to search for said drug in the 'groups' section of a popular internet search engine and take it from there.

I've also been taking it for six months or so, at 50mg, along with other meds. no real side effects, but at the same time, this has been the worst six months of my life, complete with suicidality, so i can't say it's a miracle.

Its got about a 60-70% success rate I think. (rough estimate) Some in the group have discontinued its use. So about the same as with other drugs.

My opinion is that its not quite as effective for depression and GAD as an SSRI like citalopram was (in my case), but the lack of side effects makes up for that. I am functional and making it to work etc. But I cant say Im particularly happy in general, and still have some symptoms of GAD and so my life is pretty dull and restricted.

I also have some long term gastritis problems that seem to be caused by SSRI's to begin with. Since stopping them I have found that a lot of other antidperessants including agomelatine still irritate my stomach. I dont know of anyone else with this problem however.

Overall I'd say agomelatine has the potential to help when used alone or in combination with other drugs. If your looking for minimal side effects its definetly worth a look, but as usual its not the 'perfect antidepressant' we're all looking for.

For the time being its the right drug for me.

I dont know about the US, but it is due to be launched in the UK on 1 June.

You can see the website advertising it here, it contains all the informaiton about it. I dont know how successful it is going to be with the need for a liver function test before it is prescribed then I beleive at 12 weeks then regularly afterwards.

Valdoxan resynchronizes circadian rhythms in animal models of circadian rhythm disruption.??? what does that mean?

Valdoxan increases noradrenaline and dopamine release specifically in the frontal cortex and has no influence on the extracellular levels of serotonin.

In humans, Valdoxan has positive phase shifting properties; it induces a phase advance of sleep, body temperature decline and melatonin onset. phase shifting????

> Valdoxan resynchronizes circadian rhythms in animal models of circadian rhythm disruption.??? what does that mean?

probably to do with melatonin. taking melatonin in the evening entrains the body to be tired in the night, and awake during the day.

> Valdoxan increases noradrenaline and dopamine release specifically in the frontal cortex and has no influence on the extracellular levels of serotonin.

due to 5ht2c antagonism

> In humans, Valdoxan has positive phase shifting properties; it induces a phase advance of sleep, body temperature decline and melatonin onset. phase shifting????

geting sleep cycle normal.

It has a sedative effect in the evening and (surprisingly) an activating effect in the day.

I wondered the same thing. Reading Servier's site,it doesn't sound like they're necessarily contraindicated (it says, I recall, that agomelatine doesn't effect extra-cellar serotonin). I've tried emailing them questions before and not gotten a response. Perhaps someone in Europe could do that.
