It's an amino acid found primarily in tea, which is thought to counteract the potential anxiety that the caffeine in tea can cause. In Japan, apparently it's added to all sorts of products (soft drinks, gum, etc.). There was a study in the news recently which suggested that L-theanine may even be an immune system booster.

I've found L-theanine's effects to be VERY mild,
even at doses as high as 500 mg. It might be nice for someone who is a bit stressed out, but it barely touches serious anxiety problems. This is just my experience, of course - and I do like having it around to take from time to time.
