Dimethylfumarate for depression/psychosis?

Posted by Brot on April 24, 2013, at 6:18:04

Dimethylfumarate was "newly" approved for multiple sclerosis. it also works on many chronic inflammation related skin deseases.

because of following study i gave it for me a try. for sizoaffective disorder, depression type.

Dimethylfumarate induces immunosuppression via glutathione depletion and subsequent induction of heme oxygenase 1.

Abstract
A mixture of different fumaric acid esters (FAE) is established for systemic therapy of psoriasis, a frequent inflammatory skin disease. The main active compound of FAE, however, has not been identified so far, and the mechanisms of activity are only partially understood. We analyzed the impact of FAE on in vitro immune function and aimed to gain knowledge about the mode of action. Dimethylfumarate (DMF) and diethylfumarate (DEF), but not fumaric acid, methylhydrogenfumarate and ethylhydrogenfumarate, exhibited potent depression of inflammatory cytokine secretion (e.g., tumor necrosis factoralpha, IL-12, and IFNgamma) in activated human peripheral blood mononuclear cells. Moreover, solely DMF and DEF inhibited alloreactive T-cell proliferation in mixed leukocyte reaction. Interestingly, these immunosuppressive effects were accompanied by the strong induction of the anti-inflammatory stress protein heme oxygenase 1 (HO-1). Supplementation with exogenous glutathione (GSH), which is known to bind DMF, prevented both HO-1 induction as well as the anti-inflammatory effects of DMF. Moreover, inhibition of HO-1 activity restored the diminished IL-12 and IFNgamma production after FAE treatment. These results suggest that DMF acts as active compound within the FAE mixture and at least partially mediates its immunomodulatory activity by the induction of the anti-inflammatory stress protein HO-1 ascribed to the functional depletion of reduced GSH.

any else tried it for mental stuff?

I cant say much. been on it about a week. but it seems promising.

bought the substance via a chemical supplier here, because it isnt available as a drug by now.

TECFIDERA is approved in the US for multiple sclerosis.

I've checked on the internet and found DMF dirt cheap at one place, did you get it medical grade someplace?

Have you tried MAOIs?

Good Luck!

Well, DMF is not a regulated or controlled substance, it has many non-medical uses so you can buy it in bulk at a chemical supplier like you could order Zinc or Magnesium in bulk, at least this is what I assume Brot meant with buying it via a chemical supplier.

well sadly i got a bit sick from it and stopped it. didnt get beyond 100mg bid. i dont know. its strange. it felt like an infection with fever, but no fever present. i was dizzy, weak and very tired for almost a week. i dunno, maybe it triggered some diabetic like condition.

but generally it felt like a smooth neuroleptic/mood stabiliser. my remaining voices which i hear from time to time at the evening where gone. my brain felt a lot fitter, clearer.
I didnt feel any mood brightening effect.

bt i guess i will try it again in a few weeks when my blood lab results are here and are fine. lets see if it makes me "sick" again or it was just a coincidence.
