Amisulpride users - dose question ( for dysthymia)

Hello,

I know that the "standard" dose for depression/dysthymia is 50 mg. I'm just wondering if most people find that dose works well for them. And do you take other meds with the amisulpride?

For me, my experience was that 50 mg was very sedating - so I'm considering trying it again at a lower dose...

Does anyone else take it at 25 mg? ...or even 12.5 mg? Just wondering... Thanks -

I take 50mg amisulpride/day for depression/dysthymia. It has a very mild beneficial effect. I think this has faded over time. I was prescribed 200mg in hospital for psychosis, and I know someone who is taking 800mg for Schizophrenia. Amisulpride is probably the least sedating out of all the antipsychotics. I don't think it has any affinity of histamine receptors and it doesn't make me feel drowsy. Sometimes elevated levels of dopamine makes people feel sleepy though, so it might be related to the therapeutic effect.

I've taken amisulpride, usually in doses of 50 mg, but occasionally 100 mg. Due to cross tolerance of the drug with sulpiride, and possibly methylphenidate, I haven't experienced any conspicuous effects of amisulpride. My impression of both sulpiride and amisulpride after tolerance developed to their effects, is that they have subtle stimulating - wakefulness- and alertness-promoting - effects. Those subtle effects are in contrast to my initial response to sulpiride: a stimulant effect more powerful than ever experience with methylphenidate - the improvement of motivation and initiative was particularly impressive.

I'm surprised that you're finding it sedating at such low doses, especially since you haven't reported the same effect with sulpiride. You may wish to evaluate it as a sleeping aid at higher doses, as well as testing whether minimal doses - 25 mg and less - are more stimulating.

Enhanced dopaminergic function can be stimulating and wakefulness-enhancing, but sometimes calming or perhaps even sedating. The calming effects are often observed in hyperactive children treated with stimulants. I noticed it myself also with my first doses of phenylethylamine (PEA), which had such a remarkably calming effect that I could have fallen asleep, had I wanted to.

Wikipedia also reports that amisulpride has affinity to the GHB receptor, but that seems unlikely to be of significance, since the sedative properties of GHB itself are rather more clearly mediated by the GABA-B receptor, to which amisulpride has no known affinity. See:

Just thought I should add that I do get the sedating effect with sulpiride too, at doses as low as 300 mg. (possibly even a bit lower)
