Moclobemide.....advice please

Posted by jasmineneroli on March 22, 2004, at 0:43:32

Hey everyone, I've not been here for a while...was doing pretty good and very busy with a new lifestyle....but now my GAD has worsened and I've been crashing somewhat. So my Pdoc has added Manerix (Moclobemide) a RIMA, to my daily Klonopin.
Anyone have any experience with his drug at all? Successes/horror stories? I can't find out much about it. My PDoc seems to love it, he's been trying to get me to take it for months. And I've felt so out of control lately, that I've agreed to a trial.
Thanks you guys.
Jas

I was on this for awhile--two years. It caused hormonal problems for me. It seemed pretty good on SEs aside from that. It didn't behave like a stimulant, and didn't cause sedation. Please write when you have taken it and say if you think it helps? I hope so for you, best of luck.

I tried Moclobemide for bad depression.I had no side effects, but it didn't do a thing for me.In fact,I think it made my depression worse. This stuff was like poison.

I have two "personal" experiences...I have tried the drug and it didn't do anything for me...neither positive nor negative effects.

However, I have a friend who has taken it and it worked extremely well for her.

It was for me too.

Hey Scott,
This is very interesting. Haven't you had experience with Parnate or Nardil? [I don't recall exactly]. Perhaps the people who do well on a non-reversible MAO inhibitor like these do poorly on the reversible kinds like Moclobemide.

Both Parnate and Nardil afford me transient relief. I would consider myself a partial responder to both of them. I also experienced a partial response to clorgyline, an investigational MAOI that is used now only for animal experiments and assays.

Your idea is very interesting. In psychiatry, I think it is important to look for associations where ever they can be found. However, my initial reaction is to say that there is probably no negative association between an individual's response to reversible versus irreversible MAOIs. I think it is productive to acknowledge that the reversibles are generally not as potent as the irreversibles with regard to the treatment of severe depression. It is their reversibility that most likely renders them less effective.

Moclobemide is a rather weird drug, though. It actually binds rather tightly to the enzyme compared to the other RIMAs (Reversible Inhibitors of Monoamine oxidase type A) that have been investigated. It also has an incredibly short half-life - about an hour if I recall. I think, perhaps, that it is the rapid drop in post-dose blood levels and thus the brevity of exposure that contributes to its observed reversibility.

Thanks for your responses...a mixed reaction for sure. I know it's always down to the individual, re responses to drugs, but it's good to get feed back.
Scott, your hypothesis is interesting.
I'm wondering if Manerix is really going to be any use for for my GAD, since it seems mainly to be an anti-depressant. I know most SSRI's have anti-anxiety effects in addition to anti-depressant activity......been there, done it. But they ended up making me depressed, when I didn't have depression to begin with!!! I'm wondering if Manerix (or any RIMA/MAOI) will do the same??? Any thoughts?
It is true that my anxiety/rages/out of control feelings sound like a bit of depression now, but I honestly feel, that it's something else. I can never focus, get scattered and forgetful, my creativity/thoughts are running wild and I get very obsessive, but with extreme irritability and loss of control (or feeling that way, anyway). I think it's like adrenaline overload and my "cerebral brain" cannot take over. My Pdoc insists I can't possibly have ADD or ADHD (cuz that's how I feel sometimes) since I did not have it as a kid.
Isn't there any kind of "adult onset ADD"?
The only thing that has ever helped is Klonopin, and I have kept on a very low dose for almost a year. If benzo's are effective for me, what does this say about my brain chemistry? I need GABA to function?
Any ideas for alternative drugs for the above scenario? I've been on Moclobemide for a week and will be reassessed in 2 weeks. Looking at all options.
Thx
Jas
