agomelatine and mucuna pruriens: safe?

Posted by Tusco on April 12, 2011, at 14:29:51

Hi,
I am on agomelatine 50 mg per day and I would like to add on it Mucuna Pruriens. In doing so I would look for potentiating the dopaminergic effect of agomelatine to tackle some features of my depression such as low motivation and anhedonia.
Is this combination of ago + mucuna safe?
Thanks!

Wow. There sure are a lot of active drugs in that stuff. The spines apparently contain 5-ht, so if you could only get it into your brain... With so many active chemicals in that plant, I doubt anyone could predict what interactions you might get.

Agomelatine apparently depends on the liver enzymes CYP1A2 and CYP2C9 for its metabolism. So that is a potential source of interactions. If anything in mucuna pruriens inhibits those enzymes, you may have your levels of ago increased, and conversely, if ago inhibits those or some other enzymes, you may overdose on something in mucuna pruriens. Agomelatine is probably not going to kill you, but something else might.

I didn't use Mucuna in the end. My idea was to increase dopamine and noradrenaline. I eventually went for the safer option of taking L-tyrosine. which at 3500mg has done so far nothing at all to improved my depression with low motivation features. I will probably increase further l-tyrosine up to 7-8g per day and if doesn't work I will ditch agomelatine and try reboxetine with l-tyrosine.
I am pretty f*ck*d up mentally at the moment...I'd love to find something that works.

yes I tried Bupropion in the past and did't work for me. I tried also ritalin and it didn't wok too. I haven't tried other stimulants though.
I really hope that l-tyrosine can help because it's side effects free, which makes it great. Today I got to 4,5mg and finally it seemed to kick in.
I'll see if this is a real effect or a placebo.

by 'didn't nwork' do you mean you didn't feel anything, titrated to x amount but experienced too much side effect to increase, or...

at least as focus goes, i couldn't feel anything from ritalin (on the dosage my pdoc would give me) but adderall was useful (similarly to bupropion)

That's interesting. I also got no benefit from Ritalin, but got benefit from bupropion and dextroamphetamine.

I tried both Ritalin and Bupropion yeras ago and my notes are patchy. I think that I was on 300 mg of Bupropion, which didn't lift my mood or help with motivation. The side effect that I remember was that it made me eat more and it gave me cravings for sugary foods and weirdly it made me love guinness.
I added on top of bupropion ritalin which made me hyperfocused but it did not improve either motivation or depression although I don't remember the dosage and I tried it only for few days, in fact I had a limited supply.

The only weird thing is that you don't love guinness w/o :)
