agomelatine at 1 week, give up at what point?

Not noticing any effects. Maybe harder to wake up, not refreshed at all. This could just be part of my usual pattern tho.

One night I took two instead of one (i.e. 50mg), I may have noticed somewhat peaceful feelings the next day. Nothing amazing.

How much longer should I give it? It seems I can only go up to 50mg officially after 2 weeks. Given the huge amount of legal song and dance that went into getting this drug, I don't think my doctor will break the rules.

I am likely an atypical case compared to many trying the drug. I am anhedonic, can't focus or remember anything that I'm doing. Akathitic. Cannot stop moving/talking purposelessly. Brooding on negative, cannot let go of negative, cannot respond to any positive, or see anything that inspires me. Emotionally cannot handle much. Annoying to be around. Obviously won't be taking any classes this semestre.

I think the only thing that may change that is tandospirone, due to my response to buspirone after liver enzyme wipeout by nefazodone. Usually consider highdose buspirone out of the question given the D2 antagonism, and the complex tardive syndrome i never got rid of. Have no idea how i will get tandospirone. Would consider aripiprazole if it weren't so strange, and so linked to movement issues. It is not available under normal circumstances in Canada, have little or no transportation options to cross the border.

I think I need to get the focus to get ANY info on rx'ing tandospirone for my doctor, to have the least time wasted between this trial and the next. If he wants to go through this process again with another company, doubtful unless Dainippon Sumitomo just sends it and doesn't have a song and dance beyond the easily obtained government approval (like how Sanofi-Aventis just gave me amisulpride, but charged a fair amount.) I don't know if Dainippon Sumitomo has any offices in Canada tho.

I agree give it several weeks at 50 mgs. Just talk to the doctor. I am interested in how things go as I am thinking of trying it myself. Hope you find success with it and start feeling better.

What helps with anhedonia?

My first reaction to agomelatine (noticed by others) was that I was calmer and more self posessed.

Hmmmm.

The drug worked in 2 phases. from the beginning the calmness started along with the sleep improvement. After week 2 something else started which I had some difficulties with. Basically a stimulating feeling, noticeable as anxiety in the afternoon. This isn't what you are talking about, IMO. I'm a pretty anxious type, which might explain why I liked it from the start.

I don't keep a mood diary.
Today is not a good day for me; I know that.
To begin with it was great.
I dunno....I (we?) am (are?) vulnerable and I need to look after myself.
The 1001 things that flesh is heir to, well half of them we call depression, so is it any wonder that something something something.
It's only temporary.
That's how old people keep their spirits up: everything just gets worse and worse, but time gets more and more limited, which has an AD effect of its own.

for me buspirone after liver enzyme knockout by nefazodone. want to find a safe way to replicate this. hence my focus on serotonin.

i see this as a PFC/mesocortical-pathway issue in my case, the nature of my problems and my long lasting tardive movement issues seem to point here (i can go into detail, but i'll be brief, they were SSRI induced, theorised to be related to choking VTA dopaminergic projections through te mesocortical pathway to the PFC).

5-HT2c has been mentioned as being of possibly special relevance when it comes to akathisia, including aripiprazole akathisia (5-HT2c partial agonism). i'm under the impression mCPP can be especially unfun and reported to cause akathitic dysphoria in a significant number of people.

i definitely need this. no response yet. my buspirone response gave me a feeling of how livable, calm, and under my control things could be.

day 8, still no such response. i think i may have responded when i took a double dose one night.

this makes me worry a lot. especially since this effect seems to be invoking movement related terminology (RLS), and i found it very confusing. my impression was that 5-ht2c antagonism would be anxiolytic, and very pro-healthy-movement-states. i though 5-ht2c inverse agonism was being eyed for antiparkinsonian potential...

i want tandospirone.

That's not my feeling, though I know someone for whom it has.

I need to work out what it is that wrecks my sleep. I am always having this debate about last nights sleep. Was it the wine, the grape seed extract, the arthritis stuff, or any one of the other things that I take that did it. Or is it because I'm old and f*ck*d, in which case I should relax because there's nothing I can do about that.

Well, it increases available dopamine somewhere or other.

That afternoon anxiety thing that not everybody gets....I wonder if that is related to NE? There has been some inconclusive discussion of this.
