Why AMISULPRIDE works only for 2 days? WHY??

HI to everybody.
For my unipolar depression (melanchonic, anergetic, unmotivated...) no one AD i've tried have ever worked. I think i was on every AD available in my country and every time the negative sympthoms get worse (somnolence, anergia, dizzyness)...no one signal of improvement at all.
The only drug that gave me the illusion of working is Amisulpride. I was on it lot of times (50 mg/d and 25 mg/d), but everytimes it happends the same things: for the first 2 days i feel really better: energic, motivated, alert, willing to work, to ride, to meet people...in few words to live.
But since the 3rd day every good effect vanished and i feel little dizzy, heavy eye, somnoleced, slow moving, amotivated. Why it happends?
Could anyone try to explain to this strange effect?
Parhaps it begin to work like an antipsichotic, reducing dopamine instead of increase it?
I have never went up the 1st week, should i stay more time on it to feel if s/e will reduce?
Any help will be greatly appriciate!!!!!!

Hello

I haven't tried it yet but I read many places that it poops out only after 4-5 months.

You notice improvement the first 2 days cause your body is just allowing it do what it does. After some time (your case 2 days) the body starts to fight back the drug. However I think that if you continue taking it it will kick in after 2-3 weeks and the initial improvement will return gradually.

Sounds great that it works this way. I consider it for Social Anxiety and sounds that it can be taken AS NEEDED and still helpful.

I will update about my experience with it if I get it on Monday from my doctor. Not sure if he would rx an antipsychotic for Social Anxiety.

Was it easy for you to get it?

Hi Zuzu! Thanx for your share!
I think it is a miracle that a drug could work in few days, where other AD need 4 week trial!
Here in Italy, Amisulpride is easily prescribed by the "family doctors" and you can find it in every pharmacy.
The brand name is Deniban, made by Sanofi-Sintelabo (F). The drug is not given free by the National Assistence Service (Hughh!!), but the cost for a pill of 50 mg is not too much high, about 1$.

Amisulpride worked for 2 months for me. (it worked FANTASTICALLY). Then stopped. Ever since I've been experimenting with various doses - 25mg, 50mg, 100mg - none work no more. (the dose that worked during those 2 months was 50mg). I've no idea what is the mechanism of stopping Amisulpride's effects.

I think the mechansim that makes it stop lies in the fact that the dose is very low. HOWEVER, I'm by no means advocating for highr doses than 50 or 100mg.

50mg compared to the antipsychotic dose which is 400-1200mg is very low. Virtually tolerance can develop to any drug but the poop out can be significantly delayed if one takes the higher recommended doses (which for our situation will be counteractive)

Tolerance develops because the neural pathways and the structures at the synapses are VERY plastic. After some time (2 months or 4-5) the body just get it that there's something unusual here (ie continuous blockade of D2/3 autoreceptors). because of the fact that these autoreceptors are blocked the cells will build more autoreceptors ie the autoreceptors become more sensitive. Because they become more sensitive they will detect the exceeding amounts dopamine and signal to the cell STOP RELEASE of dopamine. DAMN autoreceptors!

Another possibility is that because of the more dopamine, the threshold of activating the recieving cell rises (ie it becomes less sensitive to dopamine).

I heard about someone who used it for 4-5 months then pooped out. After she resumed it 3 months later it functioned perfectly as before.

Maybe taking it for 3 weeks then 1 week without and then resuming it might prevent tolerance ???? Try it and see.

Wish me luck with the doctor on monday, hope he will RX it. I have a strategy though, I will suggest it to replace Klonopin, he will love me getting off it!

I've read your post and i'm perfectly agree with you. I think your theory should be realistic!
But i can say a little more about your question to prevent poop-out:
Your theory to use Amis cycling (3 week on 1 week off) is supported by pharmacology.
In my country is available another drug very similar to Amisulpride and is called Levosulpiride (same class Benzamydes, group Sulpyrides). I used it for months and its effects and s/e are very similar to Amisulpride.
On the paper inside box (i don't know how it is called in English) is written that the use SHOULD be discontinuated after 2-3 week for 1 week to "cleaning".
So i think it should be applicable for analogy to the amisulpride.
Actually i'm 6 days on Amis, i've noticed that dividing dosage (25 mg morning, 25 mg evening) reduce somnolence and anergy) i will stay on for at least 2 week, and after valutation of results, stopped for 1 week. But i will post during the trial in future days.

What about you? If i have understood well, you will have a rx for amis next monday for a new trial? Or i'm wrong?

Great to hear that my tentative theory is supported scientifically!

Is the other drug's active compound Sulpiride? Trade names: Dogmatil,Sulpitil or Sulparex? Yes, they are almost the same.

But why you shifted to Amisulpride?

No, I never had this medicine but hope my doctor won't be reluctant to rx it. Any ideas how to persuade him? :)
