What's going on with substance-P ?

Posted by Scott L. Schofield on November 23, 1999, at 13:58:26

What ever happened to the substance-P antagonists? There was alot of excitement over their possible use as antidepressants some months ago, but I haven't seen anything since.

Well, my current psychiatrist told me that substance P antidepressants are coming. They supposedly will be the next new med. However, I have inside information from a teaching hospital and was told by some credible psychiatry professors that there is some questioning of the efficacy of the substance-P, that the original hype about substance P was just that...hype. It may not pan out to be the great antidepressant it was made out to be during the late summer and early fall of 98.

I would like to know more myself, like to know how many more years it will be before substance P ADs are on the market. My guess is it will be 5 more years.

I would also like to know how many more years it will be before the CRF antagonists are on the market? Maybe 10 more years?

Hell, 10 more years and the neuroscientists will probably be tinkering around with the beginning stages of genetic engineering treatments for depression.

By that time, I will probably be dead. So for me, it probably wont matter anyway.

Refractory, I can relate. I have been feeling very discouraged. Another bad bout of depression, and damn, I have to check off this latest increase in meds/adding new meds as another in a line of failures. And as I start yet another strategy (lithium) I have to have a bit of hope, tho it is surely guarded hope, that it might help.
I also hold some hope in future treatments. I think we might see new AD options, but also new magnetic stimulation type options, laser options. someday I hope they can do a combo of blood tests and scans to determne a person's unique chemistry and which part of the brain is involved, and design a targeted treatment approach. for someone who has felt as hopeless and immobile as i have recently, all of this pollyannaism is absurd. But what choice do I have other than hoping?

Substance-P: The Bad... and Good News...

Merck Pharmaceuticals has seemingly shelved the substance P inhibitor for depression at this point. However all is not lost: Merck exploring other uses for the drug (eg, treatment of chemotherapy-induced emesis), and once on the market physicians may prescribe it for other indications such as depression.

Take a look at this URL: http://www.merck.com/overview/98ar/p15.htm

Because of the efficacy of tramadol (and opioids) in OCD, perhaps the substance P inhibitor will have anti-obsessional potential.

Zeke, that is some good info you have there. The info on that Merck webpage coincides with exactly what I was told last spring by some old hand psychiatrists at a teaching hospital. They told me there was some questioning of the efficacy of the Mk-869 version substance P, that it didnt work as well as they thought. So it looks like they picked up another substance P drug and cycled it back into the process.

Guess a substance P antidepressant will be out in oh, maybe 5 years? Also, dont forget other companies are working on substance P antidepressants, not just Merck. So more hope there too. Plus there are the CRF antagonists...those are what? Maybe 10 years away?
